CD56-enriched donor lymphocyte infusion (DLI) is an adoptive cell therapy consisting of donor-derived lymphocytes that have been processed to enrich for CD56-positive cells, primarily Natural Killer (NK) cells. This intervention is typically administered following allogeneic hematopoietic stem cell transplantation (HSCT), particularly in the haploidentical setting, to enhance the graft-versus-leukemia (GVL) effect and prevent disease relapse. By selecting for CD56+ cells and potentially depleting T cells, the therapy aims to provide potent anti-tumor activity while reducing the risk of graft-versus-host disease (GVHD) compared to conventional unmanipulated donor lymphocyte infusions. The enrichment is often achieved using automated immunomagnetic selection systems.
