Drug intelligence / Profile preview
CD6-ADC (also known as ATX101) is a novel antibody-drug conjugate (ADC) consisting of a monoclonal antibody targeting CD6 (specifically the high-affinity anti-human CD6 antibody UMCD6 or its humanized version ATX006) conjugated to the antimitotic payload monomethyl auristatin E (MMAE) via a cleavable valine-citrulline-p-aminobenzylcarbamate (vc-PAB) linker. CD6 is a cell surface glycoprotein expressed on T cells and natural killer (NK) cells. While CD6 is present on all T cells, only activated, pathogenic T cells vigorously divide, making them selectively susceptible to MMAE-mediated mitotic inhibition and apoptosis. CD6-ADC is being developed by Abcon Therapeutics, in collaboration with Cleveland Clinic, the University of Michigan, and Duke University Medical Center, for the treatment of T-cell-mediated disorders, including autoimmune uveitis, graft-versus-host disease (GVHD), and renal allograft rejection, as well as CD6-expressing hematological malignancies such as peripheral T-cell lymphoma (PTCL) and aggressive NK-cell leukemia/lymphoma (ANKLL).
