Overview

CD64-28z CAR-T is an experimental second-generation chimeric antigen receptor (CAR) T-cell therapy designed to target CD64 (also known as FcγRI), a high-affinity Fc receptor for IgG. CD64 is highly expressed on monocytic acute myeloid leukemia (AML) cells and monocytic leukemia stem cells (m-LSCs), which are often resistant to standard-of-care treatments like venetoclax and azacitidine. The CAR construct incorporates a CD28 costimulatory domain and a CD3ζ signaling domain. Developed by researchers at the University of Colorado, CD64-28z CAR-T has shown potent anti-leukemic activity in preclinical models, including the elimination of CD64+ leukemia cell lines and primary patient-derived AML cells in vivo and ex vivo. It is being developed as a potential treatment for relapsed or refractory monocytic AML (FAB M5 subtype).
