Overview

CD64-41BBz CAR-T is an experimental second-generation chimeric antigen receptor (CAR) T-cell therapy designed to target CD64 (FcγRI), a high-affinity IgG receptor highly expressed on monocytic cells and monocytic leukemia stem cells (m-LSCs). Developed by researchers at the University of Colorado, this therapy specifically addresses monocytic acute myeloid leukemia (AML), particularly the M5 subtype, which is often resistant to standard treatments like venetoclax and azacitidine. The construct utilizes a 4-1BB (CD137) costimulatory domain and a CD3ζ signaling domain. Preclinical studies have demonstrated that CD64-41BBz CAR-T cells effectively eliminate CD64+ leukemia cell lines and primary patient-derived AML cells, showing superior persistence compared to CD28-based constructs in murine models.
