CD7ΔCART7 is an investigational chimeric antigen receptor (CAR) T-cell therapy engineered to target the CD7 protein, which is highly expressed in T-cell acute lymphoblastic leukemia (T-ALL) and other T-cell malignancies. A critical component of its design is the genetic disruption (knockout) of the endogenous CD7 gene in the T cells, denoted by the "Δ" symbol. This modification is essential to prevent "fratricide," a phenomenon where the CAR-T cells would otherwise target and destroy each other due to the shared expression of CD7 on both the therapeutic cells and the malignant targets. By eliminating CD7 expression on the therapeutic cells, CD7ΔCART7 can effectively expand and persist to eliminate malignant CD7-positive cells. This approach has been further refined in clinical trials (e.g., BE-CAR7) using base editing to simultaneously knock out TCR and CD52, enabling the creation of "off-the-shelf" universal CAR-T products.
