Overview

CD7-DE is an Fc-optimized monoclonal antibody targeting the CD7 antigen, primarily investigated for the treatment of T-cell acute lymphoblastic leukemia (T-ALL). The antibody is engineered with two specific amino acid substitutions in its Fc-domain (S239D and I332E, referred to as the DE-variant) to enhance its binding affinity to activating Fcγ receptors, specifically FcγRIIa and FcγRIIIa. This modification significantly boosts its ability to trigger antibody-dependent cell-mediated cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP) against CD7-positive leukemic cells. CD7-DE also serves as the antibody component of the antibody-drug conjugate CD7-DE-vcMMAE, which utilizes a microtubule-disrupting payload to induce direct cytotoxicity. Preclinical studies in xenograft mouse models have demonstrated that CD7-DE possesses potent immune-mediated anti-tumor activity, particularly in subtypes like early T-cell precursor (ETP)-ALL.
