**CD7-knockout CD7-targeted CAR T cells** are a target-specific engineered T-cell therapy platform for CD7-positive hematologic malignancies. Patient-derived or donor-derived T cells are edited to delete the endogenous **CD7** gene, preventing CAR-T-cell fratricide, and are then engineered to express an anti-CD7 chimeric antigen receptor, commonly incorporating CD28 and CD3ζ signaling domains. Following intravenous administration, the edited CAR T cells recognize CD7-expressing malignant cells and mediate cytotoxic elimination. Clinical variants have been evaluated in relapsed or refractory T-cell acute lymphoblastic leukemia, T-cell lymphoblastic lymphoma, peripheral T-cell lymphoma, NK/T-cell malignancies, and CD7-positive acute myeloid leukemia.
