**CD7-RTX CAR T cells** are a form of autologous, chimeric antigen receptor (CAR) T cell therapy engineered to target the CD7 antigen, a molecule highly expressed on the surface of T cells and some malignant T cell populations. This drug uses a CD7-directed CAR and incorporates an RTX (CD20 mimotope) "safety switch", which enables rapid depletion of administered CAR T cells by subsequent infusion of rituximab if needed for safety reasons. Preclinical studies have shown that these cells can efficiently and specifically deplete CD7-positive malignant T cells both in vitro and in mouse models. Compassionate clinical use has demonstrated efficacy and tolerability in relapsed/refractory, CD7-positive Sézary syndrome, leading to rapid and durable complete remission without severe adverse events. The RTX safety switch has been validated preclinically, enabling effective removal of CAR T cells post-infusion for safety, though clinical validation is ongoing. This approach is distinct from gene-edited "fratricide-resistant" CD7 CAR T therapies targeting T cell malignancies or autoimmune diseases, as the RTX component is a unique safety modification[2][4].
