Overview

CD70 CAR NK cells are genetically modified natural killer (NK) cells engineered to express a chimeric antigen receptor (CAR) targeting the CD70 antigen. The construct typically incorporates additional modifications such as IL-15 expression (to enhance NK cell survival, proliferation, and persistence), CD70 gene knockout, and immune receptor enhancements for improved anti-tumor function. CD70 is a transmembrane protein overexpressed in various hematologic malignancies (such as B-cell lymphoma, T-cell lymphoma, and acute myeloid leukemia) and some solid tumors; it is minimally expressed on healthy tissues, making it an attractive target. These therapies are primarily being developed as off-the-shelf, allogeneic immunotherapies leveraging the innate anti-tumor activity and low risk of graft-versus-host disease of NK cells. Key innovations in some product candidates include use of iPSC-derived NK cells, multiplex gene editing, and additional cytokine support for in vivo persistence and efficacy. Both preclinical and early clinical evidence supports robust anti-tumor activity, including tumor eradication and selective lysis of malignant cells, especially in settings where alternative targets (e.g., CD19) are absent[1][2][3][5][4][6][10].
