CD70 chimeric antigen receptor natural killer (CD70 CAR-NK) cells are an investigational allogeneic cell-based immunotherapy. These cells are genetically modified to express a chimeric antigen receptor (CAR) specific for CD70 (also known as CD27L), a member of the tumor necrosis factor superfamily that is highly expressed in various cancers but has limited expression in normal tissues. The therapy is designed as an "off-the-shelf" product, often derived from umbilical cord blood or peripheral blood mononuclear cells, bypassing the need for patient-specific manufacturing. Some constructs are further engineered to secrete IL-15 to enhance the persistence, proliferation, and anti-tumor activity of the NK cells. CD70 CAR-NK cells target both the malignant cells and CD70-expressing cancer-associated fibroblasts within the tumor microenvironment, making them a potential treatment for both hematological malignancies (like multiple myeloma and AML) and solid tumors (such as renal cell carcinoma and pancreatic cancer).
