CD70-directed CAR-NK cells with IL-15 is an allogeneic, off-the-shelf chimeric antigen receptor (CAR) natural killer (NK) cell therapy engineered to target the CD70 antigen. The therapy utilizes NK cells modified via the non-viral TC Buster transposon system to express a CAR specific for CD70 and an Interleukin-15 (IL-15) expression cassette, which enhances the persistence, proliferation, and cytotoxic activity of the cells. To prevent fratricide—a phenomenon where activated NK cells kill each other due to the upregulation of CD70—CRISPR/Cas9 technology is employed to knock out the endogenous CD70 gene. This therapeutic candidate is designed to treat CD70-positive malignancies, including acute myeloid leukemia (AML), renal cell carcinoma (RCC), and various solid tumors, by targeting both tumor cells and cancer-associated fibroblasts in the tumor microenvironment.
