CD70-encoding RNA nanoparticle is a therapeutic modality consisting of messenger RNA (mRNA) that encodes the Cluster of Differentiation 70 (CD70) protein, encapsulated within a nanoparticle delivery system, typically lipid nanoparticles (LNPs). CD70 is the biological ligand for CD27, a co-stimulatory receptor expressed on T cells and NK cells. When the mRNA is delivered to and translated by target cells (such as dendritic cells or cells within the tumor microenvironment), the resulting CD70 protein is expressed on the cell surface. This expression provides a potent co-stimulatory signal (Signal 2) for T-cell activation, enhancing the magnitude and duration of the anti-tumor immune response. This approach is often used as an adjuvant strategy in cancer vaccines or as part of a multi-mRNA cocktail (e.g., TriMix) to improve the maturation and stimulatory capacity of antigen-presenting cells.
