CD70 single-CAR T cells are an experimental adoptive cellular immunotherapy designed to target CD70-expressing malignancies, such as acute myeloid leukemia (AML). These CAR T cells typically utilize the natural ligand of CD70, CD27, as the antigen-binding domain. In the context of research conducted by Memorial Sloan Kettering Cancer Center and City of Hope, these second-generation CARs incorporate a CD28 costimulatory domain and a CD3ζ signaling domain, often modified with a single active immunoreceptor tyrosine-based activation motif (1XX ITAM) to mitigate T-cell exhaustion. While effective against CD70-positive cells, they are often studied as comparators to dual-targeting CARs (e.g., Tim-3/CD70) to address antigen heterogeneity and escape in AML.
