CD70-targeted CAR T-cell with TLR2 is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy designed to treat malignancies that overexpress the CD70 antigen, such as renal cell carcinoma and certain hematological cancers. The therapy involves engineering a patient's T-cells to express a CAR that recognizes CD70, uniquely incorporating a signaling domain derived from the Toll-like receptor 2 (TLR2), specifically the Toll/interleukin-1 receptor (TIR) domain. This inclusion of TLR2 signaling is intended to provide a potent costimulatory 'third signal' that mimics innate immune activation, thereby enhancing T-cell metabolic fitness, effector function, and persistence within the immunosuppressive tumor microenvironment. By augmenting the resistance of the T-cells to exhaustion, this modality aims to overcome traditional barriers to effective CAR-T therapy in solid tumors.
