CAR27-IL15 T cells are an investigational cell therapy consisting of T cells engineered to express a chimeric antigen receptor (CAR) that utilizes the CD27 ligand to target CD70-expressing malignancies. The construct, often referred to as CAR27, incorporates the extracellular domain of the CD70 cognate receptor (CD27) as the binding moiety, typically fused to costimulatory domains like 4-1BB and the CD3-zeta signaling chain. To overcome the challenges of the immunosuppressive tumor microenvironment and improve persistence, these cells are further engineered to co-express interleukin-15 (IL-15). CD70 is highly expressed in high-risk multiple myeloma and various other cancers, correlating with poor survival. By targeting the CD70-CD27 axis, this therapy aims to provide a potent cytotoxic response, particularly in patients who have relapsed after BCMA-directed therapies, as CD70 expression remains stable in BCMA-knockout models.
