Overview

CD70-TTC is a targeted alpha therapy (TAT) consisting of a monoclonal antibody directed against the CD70 antigen, conjugated to the alpha-emitting radionuclide thorium-227 via an octadentate 3,2-hydroxypyridinone (3,2-HOPO) chelator. Developed by Bayer, this radioimmunoconjugate is designed to treat CD70-expressing malignancies, such as renal cell carcinoma (RCC) and various B-cell lymphomas. The mechanism of action involves the antibody binding to CD70 on the tumor cell surface, followed by internalization of the conjugate. The subsequent decay of thorium-227 releases high-energy, short-range alpha particles that induce lethal DNA double-strand breaks, leading to G2 cell cycle arrest and apoptosis. Preclinical studies have demonstrated that CD70-TTC maintains high binding affinity and exhibits potent, specific anti-tumor activity with a favorable safety profile in xenograft models.
