CD70CAR-IL-12 T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy engineered to target the CD70 antigen, which is highly expressed in acute myeloid leukemia (AML) and other malignancies. These cells are modified to express Interleukin-12 (IL-12), a potent pro-inflammatory cytokine, either constitutively or through a synthetic TGFβ-sensing circuit. The inclusion of IL-12 is intended to enhance T-cell proliferation, persistence, and cytotoxicity while remodeling the immunosuppressive tumor microenvironment (TME). Preclinical studies have utilized a bidirectional gene circuit design to minimize baseline "leaky" IL-12 expression while maintaining robust CAR activity. This asset has been evaluated both as a standalone therapy and as a foundational component of the more complex MODAL (MOdular Design with Asymmetric Logic) CAR-T platform, which further incorporates constitutive IL-18 expression to fully overcome TGFβ-mediated suppression.
