Drug intelligence / Profile preview
CD71 Centyrin–IFIH1 siRNA conjugate is a preclinical-stage investigational biologic-oligonucleotide conjugate developed by Aro Biotherapeutics for the treatment of Type 1 Diabetes (T1D). The drug consists of a CD71-targeting Centyrin protein (a small ~10 kDa engineered fibronectin type III domain protein) conjugated to a short-interfering RNA (siRNA) payload that targets IFIH1 mRNA, which encodes the intracellular innate immune sensor protein MDA5. By targeting CD71 (transferrin receptor), the conjugate achieves cell-specific delivery to immune cells and pancreatic cells, enabling extrahepatic delivery of the siRNA. In preclinical studies, the conjugate demonstrated efficient uptake and robust knockdown of IFIH1 in primary human dendritic cells, attenuating Type 1 interferon production. In a Non-Obese Diabetic (NOD) mouse model of T1D, it reduced blood glucose levels, delayed disease onset, and improved survival.
