Drug intelligence / Profile preview
CD73-overexpressing adipose-derived stem cells (CD73+ADSCs) are genetically engineered mesenchymal stem cells designed to overexpress ecto-5'-nucleotidase (CD73). CD73 is the rate-limiting enzyme that catalyzes the conversion of extracellular adenosine monophosphate (AMP) to adenosine, a key immunoregulatory and tissue-protective purinergic signaling molecule. By enhancing extracellular adenosine production, these engineered stem cells activate adenosine receptor signaling (specifically A2A and A2B receptors), which suppresses inflammatory pathways (such as NF-κB and NLRP3 inflammasome activation), promotes macrophage polarization toward an anti-inflammatory M2 phenotype, and stimulates autophagy and tissue regeneration. This cell therapy is being investigated in preclinical models for the treatment of neurogenic bladder and tissue repair.
