CD79b-targeted CARCIK cells armored with IL-18 are a novel form of chimeric antigen receptor (CAR) cell therapy designed for the treatment of B-cell lymphomas, particularly relapsed or refractory non-Hodgkin lymphoma. These engineered immune effector cells combine several advanced features: - They are based on cytokine-induced killer (CIK) T cells that have been genetically modified to express a chimeric antigen receptor targeting the B-cell surface protein CD79b. - The construct is "armored" by engineering the CIK-CAR-Ts to secrete interleukin 18 (IL-18), a proinflammatory cytokine known to enhance anti-tumor activity and proliferation of immune effector cells within the tumor microenvironment. This approach aims to overcome resistance seen in patients who relapse after standard anti-CD19 or anti-CD22 CAR T cell therapies, as CD79b remains highly expressed in most B-cell non-Hodgkin lymphomas even after such relapses. Preclinical studies demonstrate improved cytotoxicity, cytokine release, and proliferation compared to conventional approaches[1][3]. Early-phase clinical data suggest feasibility and safety for use in heavily pretreated lymphoma patients[1].
