CD7CAR is a second-generation chimeric antigen receptor (CAR) T-cell therapy designed to target the CD7 antigen, a cell surface glycoprotein highly expressed in T-cell malignancies. Developed by Hebei Senlang Biotechnology in collaboration with the Beijing Lu Daopei Institute of Hematology, the therapy utilizes autologous or donor-derived T cells engineered via lentiviral transduction. The CAR construct features an anti-CD7 single-chain variable fragment (scFv), an IgG4 hinge region, a CD28 transmembrane domain, and intracellular signaling domains from 4-1BB and CD3ζ. Additionally, it incorporates a truncated EGFR (tEGFR) sequence as a selection marker or potential safety switch. Clinical trials (NCT04572308, NCT04916860) have demonstrated high rates of minimal residual disease (MRD)-negative complete remission in patients with relapsed or refractory T-cell acute lymphoblastic leukemia (T-ALL) and T-cell lymphoblastic lymphoma (T-LBL), often serving as a bridge to allogeneic hematopoietic stem cell transplantation.
