CD8+ and CD4+ Donor Memory T-cells-expressing HA1-Specific TCR is an investigational adoptive cell therapy developed by the Fred Hutchinson Cancer Center. It consists of donor-derived CD4+ and CD8+ memory T cells that have been genetically engineered using a viral vector to express a T-cell receptor (TCR) specific for the minor histocompatibility antigen HA-1 (presented by HLA-A*02:01). HA-1 is a lineage-specific antigen primarily expressed on hematopoietic cells, including leukemic blasts, but not on non-hematopoietic tissues, which minimizes the risk of off-target graft-versus-host disease (GVHD). By utilizing both CD4+ and CD8+ memory T cell subsets, the therapy aims to provide direct cytotoxic activity against leukemia cells while ensuring long-term persistence and helper function. It is primarily being evaluated for the treatment of patients with recurrent or refractory acute myeloid leukemia (AML) or acute lymphoblastic leukemia (ALL) following allogeneic hematopoietic stem cell transplantation (HCT).
