CD8 antibodies are monoclonal antibodies that specifically target the CD8 (cluster of differentiation 8) glycoprotein, a co-receptor primarily expressed on the surface of cytotoxic T lymphocytes. In clinical and preclinical settings, these antibodies serve two primary functions: as imaging tracers and as T-cell depleting agents. For diagnostic purposes, radiolabeled CD8-specific antibody fragments (such as 89Zr-ZED88082A) are used in immuno-PET imaging to non-invasively quantify tumor-infiltrating CD8+ T cells, providing a "hot" vs. "cold" tumor immunophenotype that can predict response to immunotherapies like PD-1/PD-L1 inhibitors. In research, anti-CD8 antibodies are frequently used to deplete CD8+ T cells in vivo to investigate their functional contribution to anti-tumor immunity, viral clearance, or autoimmune pathogenesis. Various clones and formats, including Fc-silent versions to avoid effector function during imaging, have been developed by companies such as Genentech and Roche.
