CD8 aptamer-siRNA refers to a class of chimeric oligonucleotides (aptamer-siRNA chimeras, or AsiCs) designed for the targeted delivery of small interfering RNA (siRNA) payloads specifically to CD8+ T cells. The aptamer portion of the chimera binds with high affinity to the CD8 cell surface receptor, facilitating receptor-mediated internalization of the conjugate. Once inside the cell, the siRNA payload is processed to silence specific target genes. Different variations of this platform have been developed for distinct therapeutic purposes: silencing RGS1 (regulator of G-protein signaling 1) to enhance CD8+ T cell recruitment and anti-tumor immunity in breast cancer; silencing granulysin (GNLY) to suppress cytotoxic T lymphocyte-mediated cytotoxicity in Stevens-Johnson syndrome/toxic epidermal necrolysis (SJS/TEN) and graft-versus-host disease (GVHD); and silencing mTORC1 to promote memory T cell differentiation.
