CD8-depleted donor lymphocytes is an investigational adoptive cellular therapy developed by the Jonsson Comprehensive Cancer Center at UCLA. The therapy involves the infusion of donor lymphocytes that have been selectively depleted of CD8+ T cells using CD8-High Density Microparticle (CD8-HDM) technology, typically achieving a depletion level of greater than 95%. This approach is designed for patients with chronic myeloid leukemia (CML) who have undergone nonmyeloablative, HLA-identical sibling peripheral blood stem cell transplantation. The primary therapeutic rationale is to minimize the risk of graft-versus-host disease (GVHD)—of which CD8+ cytotoxic T cells are major effectors—while preserving the CD4+ T cell population to maintain the graft-versus-leukemia (GVL) effect and promote stable hematopoietic chimerism.
