Drug intelligence / Profile preview
CD8-targeted ADA-1 mRNA lipid nanoparticles (CD8-ADA-LNPs) are preclinical-stage, targeted lipid nanoparticles designed to selectively deliver mRNA encoding adenosine deaminase-1 (ADA-1) to CD8+ T cells. Developed by researchers at Drexel University, the University of Pennsylvania, and collaborating institutions, this therapeutic approach aims to address the progressive dysfunction of HIV-specific CD8+ T cells in people living with HIV (PLWH). In PLWH, the adenosine-adenosine deaminase-1 axis is dysregulated, with repression of the ADA locus and heightened sensitivity to adenosine-mediated immunosuppression. By selectively delivering ADA-1 mRNA to CD8+ T cells, CD8-ADA-LNPs restore ADA-1 expression, degrade excess extracellular adenosine, and enhance antigen-specific CD8+ T-cell activation and function.
