The CD80-transfected MDA-MB-231 vaccine is an experimental allogeneic whole-cell cancer vaccine developed for the treatment of metastatic breast cancer. It consists of cells from the MDA-MB-231 breast cancer cell line (which is HLA-A2 and HER2/neu positive) that have been genetically modified to express the human costimulatory molecule CD80 (B7-1). The mechanism involves providing the mandatory 'Signal 2' (co-stimulation) for T-cell activation directly on the tumor cells, which helps overcome the immunological tolerance or anergy often induced by tumors that present antigens without co-stimulatory signals. By expressing CD80, these cells can effectively activate T cells through the CD28 receptor, promoting a systemic immune response against a variety of tumor-associated antigens present on the cell line, such as HER2, MUC1, and p53. In clinical trials, the vaccine was often administered alongside adjuvants like GM-CSF or BCG to further stimulate dendritic cell activity and enhance the overall immune response.
