CD81 blocking antibodies are a class of monoclonal antibodies designed to bind to the large extracellular loop (LEL) of the CD81 tetraspanin protein. CD81 is a critical host factor for the entry of the Hepatitis C Virus (HCV) into hepatocytes, serving as a co-receptor that interacts with the viral E2 glycoprotein. By blocking this interaction, these antibodies prevent viral attachment and entry, making them potential therapeutic candidates for preventing HCV reinfection, particularly in patients undergoing liver transplantation. Beyond virology, CD81 is implicated in tumor progression, cell migration, and the entry of malaria parasites into the liver, leading to research into its use in oncology and infectious disease prophylaxis.
