CD84-CD3 bispecific antibody is an experimental bispecific T-cell engager (BiTE) developed for the treatment of acute myeloid leukemia (AML). The therapeutic is designed to simultaneously target CD84 (also known as SLAMF5), a signaling lymphocytic activation molecule family member that is highly expressed on AML blasts and leukemia stem cells, and the CD3 subunit of the T-cell receptor complex. By bridging these two targets, the antibody facilitates the formation of an immunological synapse, redirecting T-cell-mediated cytotoxic activity specifically against CD84-positive leukemic cells. Preclinical studies conducted by Shanghai Symray Biopharma have demonstrated that lead candidates, such as #24 and #26, achieve potent anti-leukemic activity and prolonged survival in xenograft and patient-derived models. Notably, CD84 expression is minimal on normal hematopoietic stem cells (HSCs), potentially offering a wider therapeutic window and reduced myelotoxicity compared to agents targeting CD33 or CD123.
