CD84-CD3 BiTE #26 is a bispecific T-cell engager (BiTE) designed for the treatment of acute myeloid leukemia (AML). It is engineered to simultaneously bind to CD84, a cell surface receptor highly expressed on AML blasts and leukemia stem cells, and the CD3 complex on T cells. By bridging these two cell types, the antibody induces MHC-independent T-cell activation and redirected cytotoxicity against leukemic cells. CD84 is considered a promising target in AML because it is minimally expressed on normal hematopoietic stem cells (HSCs), potentially offering a wider therapeutic window compared to targets like CD33 or CD123. Preclinical studies conducted by Shanghai Symray Biopharma and Zhejiang University demonstrated that CD84-CD3 BiTE #26 triggers potent T-cell mediated killing of AML cell lines and patient-derived blasts, accompanied by the release of cytokines such as IFN-γ, TNF-α, and IL-2, while sparing healthy donor cells.
