Overview

CD84.02 CAR T-cells is a second-generation chimeric antigen receptor (CAR) T-cell therapy being developed by Gyala Therapeutics S.L. for the treatment of hematological malignancies, specifically relapsed or refractory acute myeloid leukemia (AML) and T-cell acute lymphoblastic leukemia (T-ALL). The therapy targets CD84 (also known as SLAMF5), a signaling lymphocytic activation molecule family member that is overexpressed on AML and T-ALL cells but shows limited expression on healthy hematopoietic stem progenitor cells (HSPCs), potentially offering a wider therapeutic window than other myeloid targets. The CD84.02 construct is engineered with a murine single-chain variable fragment (scFv) in a VLVH configuration, a CD8α hinge and transmembrane domain, a 4-1BB costimulatory domain, and a CD3ζ signaling domain. Preclinical data demonstrated significant anti-leukemic activity in vitro and in vivo, leading to the initiation of a Phase I/II clinical trial.
