CD84.03 CAR T-cells are a second-generation chimeric antigen receptor (CAR) T-cell therapy targeting CD84 (also known as SLAMF5), an immunoreceptor overexpressed in acute myeloid leukemia (AML) and T-cell acute lymphoblastic leukemia (T-ALL). Developed by Gyala Therapeutics, a spin-off of the Hospital Clínic of Barcelona, the construct features a murine single-chain variable fragment (scFv) in a VHVL configuration, a CD8α hinge and transmembrane domain, a 4-1BB costimulatory domain, and a CD3ζ signaling domain. Preclinical studies have demonstrated its ability to control tumor progression and improve survival in AML and T-ALL mouse models. It is intended as a bridge therapy to allogeneic stem cell transplantation due to potential myelotoxicity.
