CD84.04 CAR T-cells is an investigational second-generation chimeric antigen receptor (CAR) T-cell therapy targeting the CD84 antigen (also known as SLAMF5). Developed by Gyala Therapeutics, a spin-off of the Hospital Clínic of Barcelona, this therapy utilizes a murine single-chain variable fragment (scFv) in a VHVL configuration, combined with a CD8α hinge and transmembrane domain, a 4-1BB costimulatory domain, and a CD3ζ signaling domain. CD84 is highly expressed on leukemic blasts in acute myeloid leukemia (AML) and T-cell acute lymphoblastic leukemia (T-ALL), while showing limited expression on healthy hematopoietic stem progenitor cells, potentially reducing on-target/off-tumor toxicity. Preclinical studies have demonstrated potent cytotoxicity against AML and T-ALL cells, and the therapy is entering Phase I/II clinical trials for relapsed or refractory cases.
