CD84.05 CAR T-cells are an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target CD84 (also known as SLAMF5), a cell surface protein overexpressed in various hematological malignancies. The CD84.05 construct utilizes a murine-derived single-chain variable fragment (scFv) in a VHVL configuration, paired with a CD8α hinge and transmembrane domain, a 4-1BB costimulatory domain, and a CD3ζ signaling domain. Preclinical studies have demonstrated that these CAR T-cells exhibit specific cytotoxic activity, proliferation, and cytokine secretion when exposed to CD84-positive leukemic cells, including those from acute myeloid leukemia (AML) and T-cell acute lymphoblastic leukemia (T-ALL). Developed by researchers at Hospital Clínic de Barcelona and Gyala Therapeutics, the therapy is being investigated as a potential treatment for relapsed or refractory myeloid and lymphoid malignancies, potentially serving as a bridge to allogeneic hematopoietic stem cell transplantation.
