CD84.06 CAR T-cells is an investigational second-generation chimeric antigen receptor (CAR) T-cell therapy targeting the CD84 antigen (also known as SLAMF5). Developed by Gyala Therapeutics, this therapy is designed for the treatment of acute myeloid leukemia (AML) and T-cell acute lymphoblastic leukemia (T-ALL). The CD84.06 construct features a murine-derived single-chain variable fragment (scFv) in a VHVL orientation, a CD8α hinge and transmembrane domain, a 4-1BB (CD137) costimulatory domain, and a CD3ζ signaling domain. CD84 is an immunoreceptor of the SLAM family that is overexpressed on leukemic blasts while showing limited expression on healthy hematopoietic stem progenitor cells (HSPCs), potentially offering a therapeutic window to minimize on-target/off-tumor myelotoxicity. Preclinical studies have demonstrated that CD84.06 CAR T-cells exhibit potent cytotoxic activity, proliferate, and secrete cytokines upon recognition of CD84-positive tumor cells.
