Overview

CD85d CAR-T cells are an experimental adoptive cellular immunotherapy designed to target CD85d (also known as ILT4 or LILRB2), a member of the leukocyte immunoglobulin-like receptor family. CD85d was identified through multi-omic surfaceome mapping as being highly expressed across various acute myeloid leukemia (AML) phenotypes while remaining largely absent from normal hematopoietic stem and progenitor cells (HSPCs) and vital non-hematopoietic tissues. These CAR-T cells are engineered to selectively recognize and eliminate CD85d-positive leukemic blasts, potentially offering a safer alternative to therapies targeting CD33 or CD123, which often cause on-target off-leukemia toxicity due to expression on healthy HSPCs. The therapy is currently in preclinical development at Washington University in Saint Louis.
