CD86-Fc-NKG2a is a preclinical-stage recombinant fusion protein developed by Shattuck Labs using its proprietary Agonist Redirected Checkpoint (ARC) platform. The molecule is designed to enhance anti-tumor immunity by simultaneously engaging the costimulatory ligand CD86 and the inhibitory checkpoint receptor NKG2A. CD86 (B7-2) typically interacts with CD28 to provide essential secondary signals for T-cell activation, while NKG2A is an inhibitory receptor found on Natural Killer (NK) cells and cytotoxic T cells that binds to HLA-E on tumor cells to suppress immune responses. By combining these two domains into a single hexameric fusion protein, CD86-Fc-NKG2a aims to provide potent costimulation while relieving checkpoint inhibition, thereby bridging innate and adaptive immunity to combat various cancers.
