Overview

CD93-BBz CAR T cells are a second-generation chimeric antigen receptor (CAR) T-cell therapy targeting CD93, a cell surface glycoprotein highly expressed in acute myeloid leukemia (AML) and on leukemia stem cells (LSCs). The construct incorporates a humanized single-chain variable fragment (scFv) derived from the F11 antibody clone, fused to a 4-1BB (CD137) costimulatory domain and a CD3ζ signaling domain. Preclinical studies conducted at Stanford University demonstrated potent anti-leukemic efficacy in vitro and in patient-derived xenograft (PDX) models. However, research also identified significant on-target, off-tumor toxicity due to CD93 expression on endothelial cells, suggesting that clinical application may require advanced engineering strategies, such as logic-gated or combinatorial targeting, to mitigate vascular damage.
