Overview

CD96-CAR T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target CD96 (also known as Tactile), a cell surface marker highly expressed on leukemia stem cells (LSCs) and blasts in acute myeloid leukemia (AML), while being absent on normal hematopoietic stem/progenitor cells (HSPCs). Developed to address the scarcity of LSC-specific antigens, these CAR-T cells typically utilize a CD28 transmembrane and costimulatory domain (CD96-28z) to enhance proliferation and cytotoxicity. To overcome low antigen density in some AML cases, they may be combined with a CD33-targeted chimeric costimulatory receptor (CCR). Preclinical studies indicate potent anti-leukemic activity and prolonged survival in animal models without significant toxicity to normal hematopoiesis.
