CDC-DXd is a non-targeting control drug conjugate (CDC) used in preclinical oncology research to evaluate the specificity and efficacy of antibody-drug conjugates (ADCs) targeting oncofetal chondroitin sulfate (ofCS). It consists of an irrelevant albumin-binding domain (ABD) protein scaffold (ABDwt-SpyC-2Cys) conjugated to the cytotoxic payload Deruxtecan (DXd) via a cathepsin-cleavable glycine-glycine-phenylalanine-glycine (ggfg) linker. CDC-DXd serves as a negative control to distinguish between the targeted anti-tumor activity of Vartumab-DXd and non-specific effects such as systemic payload release or bystander killing. In experimental models of melanoma and lung cancer, CDC-DXd typically demonstrates minimal anti-tumor activity compared to the targeted Vartumab-DXd, thereby validating the therapeutic window and targeting mechanism of the ofCS-directed ADC.
