Overview

CDC-MMAE is a non-targeting control drug conjugate (CDC) used in preclinical oncology research to evaluate the specificity and therapeutic index of antibody-drug conjugates (ADCs). It consists of an irrelevant protein scaffold, specifically an albumin-binding domain (ABD) fusion protein, conjugated to the cytotoxic payload monomethyl auristatin E (MMAE) via a cathepsin-cleavable valine-citrulline (vc) linker. In studies involving Vartumab-based ADCs, CDC-MMAE serves as a negative control to distinguish between target-mediated anti-tumor activity and non-specific toxicity or bystander effects. It typically features a drug-to-antibody ratio (DAR) of approximately 1.9 and is used to validate that the efficacy of targeted ADCs is driven by specific binding to antigens like oncofetal chondroitin sulfate (ofCS).
