The cDC1 vaccine is an investigational autologous dendritic cell-based immunotherapy developed by the H. Lee Moffitt Cancer Center and Research Institute, in collaboration with Roswell Park Comprehensive Cancer Center. It is specifically designed to treat breast cancer leptomeningeal disease (LMD). The vaccine consists of type 1 conventional dendritic cells (cDC1s) that are pulsed with MHC class II peptides derived from HER2 and HER3 tumor antigens. By administering the vaccine intrathecally via an Ommaya reservoir, researchers aim to stimulate a robust Th1 CD4+ adaptive immune response directly within the cerebrospinal fluid, addressing the immune deficiency typically found in the leptomeningeal space. It is currently being evaluated in Phase 1 and Phase 2 clinical trials for patients with HER2-positive or triple-negative breast cancer LMD.
