Overview

This is a cancer peptide vaccine composed of three synthetic peptides derived from the tumor-associated antigens CDCA1 (cell division cycle associated 1), IMP-3 (insulin-like growth factor-II mRNA-binding protein 3), and LY6K (lymphocyte antigen 6 complex locus K). These antigens are frequently overexpressed in various cancers, including head and neck squamous cell carcinoma (HNSCC), esophageal squamous cell carcinoma (ESCC), non-small cell lung cancer (NSCLC), and others, but not in normal tissues. The vaccine is designed to induce cytotoxic T lymphocyte (CTL) responses specifically against tumor cells expressing these antigens. Clinical trials have shown that vaccination with this three-peptide combination can elicit robust CTL responses in HLA-A24-positive patients, which correlates with improved progression-free survival and overall survival. The mechanism of action is immunotherapeutic: the peptides are presented by HLA-A24 molecules on antigen-presenting cells, activating specific CTLs that target tumor cells expressing CDCA1, IMP-3, or LY6K[2][7][8].
