CDCA1 peptide is an investigational cancer vaccine component consisting of an HLA-A*24:02-restricted epitope derived from the cell division cycle-associated protein 1 (CDCA1), also known as NUF2. Developed by the University of Yamanashi, it is primarily targeted at patients with esophageal squamous cell carcinoma (ESCC). CDCA1 is a cancer-testis antigen that is highly and frequently expressed in ESCC but has limited expression in normal adult tissues, making it a suitable target for T-cell-based immunotherapy. The peptide works by inducing antigen-specific cytotoxic T lymphocytes (CTLs) that recognize and eliminate cancer cells presenting the CDCA1 epitope. In clinical settings, such as the STF-II Phase II trial, it has been evaluated as part of multi-peptide vaccine formulations (alongside URLC10 and KOC1) to enhance the immune response against heterogeneous tumor cell populations. This peptide was later incorporated into the five-peptide vaccine S-588410 developed by Shionogi.
