CDCP1+ mesenchymal stem cells are a specific subset of mesenchymal stem cells (MSCs) derived from umbilical cord tissues, identified through single-cell sequencing and bioinformatics analysis. These cells exhibit a stronger potential for immune negative regulation and enhanced migration ability compared to general MSCs. They are being investigated for their role in preventing and treating acute graft-versus-host disease (aGVHD), a major complication after hematopoietic stem cell transplantation. The proposed mechanism of action involves inducing the differentiation of T cells into immunosuppressive cells, potentially through the CDKN2B signaling pathway. Studies have shown that CDCP1+MSCs inhibit T cell proliferation, increase regulatory T cells (Treg), decrease other T cell subsets (Th2, Th17, Tc1, Tc2, Tc17), and alter cytokine release (decreasing TNF-α, IL-4, IL-6, IL-21 while increasing CCR2 expression).
