CDH17 nanoCAR circular RNA is a preclinical-stage in situ chimeric antigen receptor (CAR) therapy developed by Citil Pharma for the treatment of colorectal cancer. The therapeutic utilizes circular RNA (circRNA) to encode a CAR specific for Cadherin-17 (CDH17), a cell surface marker frequently overexpressed in gastrointestinal malignancies. This circRNA is encapsulated within lipid nanoparticles (LNPs) that are targeted using a nanobody against CD11b, a marker found on various myeloid and lymphoid immune subsets. By delivering the genetic instructions directly to immune cells in vivo, the therapy aims to generate CAR-NK or CAR-NKT cells within the patient's body, potentially offering a more accessible and scalable alternative to traditional ex vivo CAR-T cell manufacturing.
