CDH17-targeting ADCs represent a class of antibody-drug conjugates designed to treat gastrointestinal (GI) malignancies by targeting Cadherin-17 (CDH17), also known as liver-intestinal cadherin (LI-cadherin). CDH17 is a calcium-dependent cell adhesion protein that is highly overexpressed in colorectal, gastric, and pancreatic cancers but has restricted expression in healthy adult tissues, making it an ideal tumor-associated antigen. These conjugates typically utilize a humanized monoclonal antibody to deliver potent cytotoxic payloads—such as topoisomerase I inhibitors (e.g., MF-6 in Mabwell's 7MW4911) or microtubule-disrupting agents (e.g., MMAE in Arbele's candidate)—directly to tumor cells. Upon binding and internalization, the payload is released to induce apoptosis. Clinical development is led by candidates like 7MW4911, which is currently undergoing Phase I/II evaluation for advanced solid tumors and GI cancers.
