CDH6-ADC is an antibody-drug conjugate (ADC) consisting of a fully human IgG1 monoclonal antibody targeting Cadherin-6 (CDH6), conjugated to the tubulin-binding maytansinoid payload DM4 (ravtansine) via a cleavable sulfo-SPDB linker. CDH6 is a cell adhesion molecule frequently overexpressed in ovarian and renal cell carcinomas, while maintaining restricted expression in normal tissues. Upon binding to CDH6 on the tumor cell surface, the ADC is internalized, and the DM4 payload is released to inhibit microtubule assembly, leading to cell cycle arrest and apoptosis. Developed by Novartis, CDH6-ADC has demonstrated robust anti-tumor activity in patient-derived xenograft (PDX) models of ovarian and renal cancer, including those refractory to standard-of-care chemotherapy.
