Overview

CDI-45205 is a SARS-CoV-2 3CL (main) protease inhibitor developed as a potential broad-spectrum antiviral for COVID-19 and its variants. It was discovered through a collaboration with Kansas State University Research Foundation (KSURF) and is being advanced by Cocrystal Pharma. CDI-45205 has demonstrated potent in vitro activity against multiple SARS-CoV-2 variants (Alpha, Beta, Gamma, Delta, Omicron) and other coronaviruses, showing efficacy that surpasses the original Wuhan strain[1][3]. Additionally, it has shown preclinical antiviral activity against other viruses including noroviruses and rhinoviruses[3]. The drug has demonstrated good bioavailability in animal pharmacokinetic studies (mouse, rat, intraperitoneal), no cytotoxicity in a range of human cell lines, and a strong synergistic effect with remdesivir[1][2][3]. In vivo proof-of-concept studies have also shown efficacy in MERS-CoV-2 infected mice[1][2]. Cocrystal Pharma is developing CDI-45205 as both an intranasal/pulmonary product and, separately, a candidate for oral administration[3]. The compound is not a repurposed drug but designed specifically as a broad-spectrum antiviral targeting viral proteases[4].
