**CDK-004** is an investigational, intravenously administered exosome-based antisense oligonucleotide therapy developed by Codiak BioSciences for solid tumors involving the liver. Also called **exoASO-STAT6**, it consists of cell-derived exosomes loaded with a synthetic lipid-tagged antisense oligonucleotide directed against **STAT6**. The candidate was designed to preferentially deliver the oligonucleotide to tumor-associated myeloid cells, silence STAT6, and reprogram immunosuppressive M2-like macrophages toward a proinflammatory M1-like phenotype, thereby promoting antitumor immune activity. A first-in-human Phase 1 study in advanced hepatocellular carcinoma and liver metastases from colorectal or gastric cancer was terminated.
